چکیده

The goal of this study was to examine the effect of caffeine on motility parameters of cooling-stored ram sperm – total motility and progressive movement. Caffeine is a cyclic nucleotide phosphodiesterase inhibitor, which can affect sperm motility. The semen was kept at 4 ̊C for several days. Caffeine was added to fresh ram semen diluted in Triladyl at concentrations of 1, 2 and 4 mmol.l -1 every day of sperm storage, shortly before analysis. Sperm motility was analyzed using CASA system after 24, 48, and 72 h of storage. We confirmed the stimulated effect of caffeine given at concentrations of 1, 2, and 4 mmol.l -1 on ram sperm total motility and progressive movement. The maximum effect of caffeine was observed 30 min after addition. The most effective concentration of caffeine was 4 mmol.l -1. These results indicate that caffeine can maintain motility of ram sperm during long-term cooling-storage

P-159: Effect of Silymarin on Viability, Motility and Mitochondrial Memberane Potential of Ram Spermatozoa Treated with Aluminum Chloride

Background: Aluminum, as an environmental pollutant, can exert oxidative stress on spermatogenesis and sperm morphology. Silymarin, an effective substance extracted from Silybum marianum, is a potent antioxidant which inhibits oxidative stress. Due to the damaging effects of aluminum on the male reproductive system as well as sperm and the role of silymarin, this study was performed to investig...

P-137: Improvement in Post-Thawed Ram Sperm Quality by Adding Cysteine in Soybean Lecithin Based Extender

Background: Ram sperm have a higher polyunsaturated fatty acids and a lower cholesterol/phospholipid ratio than other species which renders the sperm much more vulnerable to oxidative damage caused by reactive oxygen species (ROS). Oxidative stress can affects the integrity of cells at both the structural and sub structural levels. Therefore, a suitable antioxidant may prevent cryo-injuries and...

Effect of Adding Coenzyme Q10 and Ellagic Acid during Cryopreservation on Post-Thaw Quality of Ram Semen

The aim of this study was to determine the effects of co-Q10 and ellagic acid on sperm parameters after the freeze–thawing of Ghezel ram sperm. Twenty ejaculates from five Ghezel rams were collected in this study. Semen samples, which were diluted with a soybean lecithin (SL) based extender containing 0.25 mM ellagic acid, 0.5 μM co-Q10, 0.25 mM ellagic acid + 0.5 μM co-Q10 and no antioxidant (...

Effect of caffeine on functions of cooling-stored ram sperm in vitro

Caffeine is a well-known sperm motility stimulator, however, its effects on cooling-stored ram semen are unknown. The aim of the study was to examine the effect of caffeine on selected motility and viability indices of cooling-stored ram spermatozoa. Sperm ejaculates from 4 rams were diluted (1:3) in a Triladyl extender. Samples were stored for 96 h at 4-5 °C in two sets. In the first set used ...

Antioxidant Effects of Thyme (Thymus vulgaris) Extract on Ram Sperm Quality during Cryopreservation

Oxidative stress during freezing-thawing reduces sperm motility, viability, membrane functions and antioxidant capacity. Thymus vulgaris has antioxidant properties due to phenolic compounds. The purpose of current study was to evaluate the effect of Thymus vulgaris extract as a natural antioxidant on post-thawed Moghani ram sperm quality. Semen samples were collected twice a week from four rams...
